The Science

A different approach to restoring response

PMJ2 is designed to act upstream of checkpoint blockade by inducing cytotoxic ER stress and DAMP signaling in tumor cells. This process is intended to promote immunogenic cell death, activate dendritic cells, and prime tumor-specific T-cell responses that may help re-sensitize resistant tumors to checkpoint inhibitors.

Turning resistant tumors into immune-responsive tumors

Overcoming resistance requires more than blocking PD-1, PD-L1, or CTLA-4 alone. PMJ2 is designed to increase cytotoxic immune-cell infiltration and help convert “cold,” resistant tumors into “hot,” more immune-responsive tumors that are better positioned to respond to checkpoint blockade.

Evidence of anti-tumor activity

In preclinical melanoma models, PMJ2 has demonstrated tumor growth suppression and selective toxicity toward tumor cells. Claredele’s data also support cytotoxic ER stress and immunogenic cell death as key components of PMJ2’s anti-tumor effect.

  • Tumor growth suppression in melanoma models by >70%.

  • Evidence of ER stress markers and apoptosis.

  • Findings consistent with immunogenic cell death.

Built to support durable immune memory

Beyond direct anti-tumor activity, PMJ2 has shown evidence of generating memory T-cell responses in preclinical melanoma models. These findings suggest the potential for a more durable immune effect, supporting the rationale for longer-lasting immunotherapy benefit.

  • Increased memory T-cell infiltration in secondary tumors.

  • Evidence consistent with durable immune activation.

  • Supports the rationale for longer-lasting immunotherapy benefit.

Designed for combination with checkpoint inhibitors

PMJ2 is being developed to enhance—not replace—checkpoint inhibitors. By restoring immune activation in resistant tumors, Claredele aims to make existing PD-1, PD-L1, and CTLA-4 therapies more effective for patients with limited options.
